dc.description.abstract[cat] La lipoproteïna lipasa (LPL) és un enzim àmpliament distribuït a l’organisme i aquest fet li confereix un rol central en el metabolisme lipídic. La seva funció principal és la hidròlisi dels triacilglicèrids de les lipoproteïnes circulants. D’aquesta hidròlisi s’alliberen àcids grassos no esterificats que seran captats pels teixits on seran utilitzats en funció del context fisiològic. L’enzim se sintetitza principalment al teixit adipós, al cor, al múscul esquelètic i a la glàndula mamària alletant. En la seva forma madura i activa, l’LPL és un dímer que és secretat per les cèl·lules parenquimals del teixit on es localitza i és transportat fins a la llum dels vasos on exercirà la seva funció. En estudis del nostre grup, es va trobar que l’LPL de cor i de plasma post-heparínic (PHP) de rata presenten isoformes del mateix pes molecular (MW) aparent però diferent punt isoelèctric (pI). Es van estudiar els possibles orígens de l’heterogeneïtat de pI d’aquestes isoformes concloent que: (i) les isoformes de l’LPL no corresponen a formes intermèdies de maduració intracel·lular i (ii) l’origen no es pot atribuir a fosforilacions de la proteïna però, en part, sí que es pot explicar per diferents graus de glicosilació. En aquesta tesi, ens vàrem proposar estudiar si l’espècie humana també presenta isoformes de pI de l’LPL. Hem comprovat per primera vegada que, tant el PHP, com la llet i el teixit adipós blanc (WAT) subcutani d’humans presenten isoformes de l’LPL. A més, hem descartat que l’origen molecular sigui degut a variacions al·lèliques o provinguin de diferents òrgans, estudiant les isoformes provinents d’un únic teixit d’un sol individu. A més, hem ampliat l’estudi de la distribució tissular de les isoformes de l’LPL en rata demostrant que, en alguns fluids i teixits de rata (llet, PHP de femella i WAT retroperitoneal), coexisteixen, juntament amb les isoformes de pI de l’LPL, isoformes de MW aparent. Hem postulat que podrien tenir el seu origen molecular en modificacions post-traduccionals, concretament, en la glicosilació. Cal destacar també en rates que el patró d’isoformes de l’LPL divergeix segons el gènere i el tipus de WAT. També, ens vam proposar avançar en la caracterització funcional de les isoformes de l’LPL d’una manera indirecta, ja que actualment no disposem d’eines per estudiar cada isoforma per separat. Així, vàrem estudiar si el patró d’isoformes de l’LPL varia en situacions fisiològiques (alletament) i patològiques (obesitat) en les quals l’activitat de l’enzim està altament regulada. Donada la impossibilitat d’obtenir glàndula mamària alletant humana, vàrem estudiar les isoformes de l’LPL en rata alletant. En aquest model hem vist que els patrons de les isoformes dels fluids i teixit estudiats (llet, PHP i glàndula mamària) divergeixen en els seus pI, MW aparents i quantitats relatives. Probablement aquestes variacions tinguin algun significat fisiològic. També hem comparat els patrons de les isoformes de l’LPL del WAT subcutani d’individus normopès i d’individus obesos. Degut a la dificultat d’obtenir WAT visceral d’humans normopès, vàrem ampliar l'estudi a dos models murins d’obesitat: induïda per dieta de cafeteria i per modificació genètica. Dels resultats concloem que en l’obesitat les isoformes de l’LPL tendeixen a tenir una major representació en la zona bàsica del rang de pH. Complementant la caracterització funcional, ens vam plantejar estudiar si les diferents isoformes presenten diferències d’activitat lipolítica. Gràcies a la posada al punt del mètode del substrat suïcida per lipases i a la seva adaptació al nostre enzim, podem concloure que totes les isoformes de l’LPL, purificada parcialment mitjançant cromatografia d’afinitat a heparina-Sepharose, són actives. L’LPL és una proteïna complexa i calen més aproximacions, tant a nivell molecular com funcional, per comprendre el sentit biològic de les seves isoformes.-
dc.description.abstract[eng] Lipoprotein lipase (LPL) is an enzyme widely distributed in the body and this gives it a central role in lipid metabolism. Its main function is the hydrolysis of triacylglycerides of circulating lipoproteins. In previous studies, it was found that LPL of rat present isoforms of the same molecular weight (MW) apparent but different isoelectric point (pI). In this thesis, we verified that the human species presents pI isoforms of LPL in: post-heparin plasma (PHP), milk and white adipose tissue (WAT). In addition, we have shown that in some fluids and tissues of rat (milk, female PHP and retroperitoneal WAT), together with the pI isoforms, apparent MW isoforms of LPL coexist. Also, we advanced in the functional characterization of the LPL isoforms in an indirect way. In this way, we studied whether the isoform pattern of LPL varies in physiological (lactation) and pathological (obesity) situations in which the activity of the enzyme is highly regulated. Given the impossibility of obtaining a human lactating mammary gland, we performed the lactation study in rats. In this model the patterns of the isoforms of the fluids and tissue studied (milk, PHP and mammary gland) differ in their pI, apparent MW and relative amounts. Probably these variations have some physiological meaning. We also compared the patterns of the LPL isoforms of the subcutaneous WAT of normo-weight and obese individuals. Due to the difficulty of obtaining visceral WAT from normal-weight humans, we extended the study to two murine models of obesity: induced by cafeteria diet and by genetic modification. We concluded that in obesity the LPL isoforms tend to have a greater representation in the basic zone of the pH range. Complementing the functional characterization, we proposed to study if the different isoforms present differences of lipolytic activity. Thanks to the development of the method of the suicide substrate for lipases, we concluded that all the isoforms of the LPL, partially purified by heparin-Sepharose affinity chromatography, are active. LPL is a complex protein and more approximations are needed to understand the biological meaning of its isoforms.-
