dc.description.abstract[cat] En plantes, els fotoreceptors que han estat millor caracteritzats són els fitocroms (phys), que capten llum roja (R, de l'anglès red), y roja llunyana (FR, de l'anglès far red). Entre tots els processos regulats pels fitocroms, la síndrome de fugida de l'ombra o SAS (de l'anglès shade avoidance syndrome) és probablement el més important en les plantes crescudes en llum. La SAS fa referència a un grup de respostes induïdes quan plantes completament desetiolades creixen en alta densitat, és a dir, en proximitat d'altres plantes veïnes. Malgrat la complexitat d'aquestes respostes, la SAS s'indueix per un sol senyal ambiental, la disminució de la raó de llum R:FR. La percepció d'aquest senyal pels fitocroms inicia una complexa xarxa transcripcional. Per identificar nous components de la SAS, ens hem centrat en un grup de gens l'expressió dels quals està ràpidament afectada per llum de baixa raó R:FR. En aquesta tesi hem caracteritzat funcionalment dos nous gens d'aquest grup: PAR1 i PAR2 (de l'anglès phy rapidly regulated), que codifiquen factors de transcripció bHLH atípics. Plantes transgèniques amb nivells reduïts d'aquests gens mostren una resposta incrementada a l'ombra simulada, mentre que plantes de sobreexpressió de PAR1 i PAR2 presenten un fenotip nan i un color verd fosc, oposat a la SAS. Aquestes dades suggereixen que PAR1 i PAR2 regulen negativament les respostes de la SAS. A més, anàlisis de micromatrius ens han permès identificar un grup de gens reprimits per PAR1 de la família dels SAUR (de l'anglès small auxin up RNAs). L'expressió d'aquests SAURs s'indueix tant per auxines com per brassinosteroides de manera dosi-dependent, però també la llum de baixa raó R:FR els indueix transitòriament. L'anàlisi d'expressió dels gens SAUR15 i SAUR68 en tractaments combinats de llum i hormones (auxines i brassinosteroides) en plantes silvestres així com en línies amb nivells alterats de PAR1 suggereixen que aquest factor podria actuar com a repressor de la transcripció d'aquests dos gens SAUR. Els resultats obtinguts en aquesta tesi suggereixen que PAR1 modularia el desenvolupament integrant senyals de llum i d'hormones.cat
dc.description.abstract[eng] The best characterized photoreceptors in plants are the phytochromes (phys), which perceive red (R) and far-red (FR) light. Among all the processes regulated by phys, the shade avoidance syndrome (SAS) is probably the most important for light-grown plants. The SAS refers to a group of responses induced when fully de-etiolated plants grow in close proximity of neighbouring vegetation. Despite the complexity of these responses, the SAS is induced by a single environmental signal, which is the decrease in the R: FR ratio. The perception of this signal by the phytochromes initiates a complex transcriptional network. In order to identify new components of the SAS, we have focused in a group of genes which expression is rapidly affected by low R: FR ratio light. We show functional data about two new genes of this group, PAR1 and PAR2 (from phy rapidly regulated), which encode atypical bHLH transcription factors. Transgenic plants with reduced levels of these genes show an increased response to simulated shade, while PAR1 and PAR2 overexpression plants show a dwarf phenotype and a dark green colour, opposite to the effect of the SAS treatment. These data suggest that PAR1 and PAR2 negatively regulate the SAS responses. Moreover, microarray analyses allow us to identify a group of genes repressed by PAR1 belonging to the SAUR (small auxin up RNAs) family. The expression of these genes is induced by auxins as well as brassinosteroids in a dose-dependent manner, but it is also transiently induced by low R:FR ratio light. The expression analysis of SAUR15 and SAUR68 genes in treatments combining light and hormones (auxins and brassinosteroids) in wild type plants as well as plants with altered PAR1 levels suggest that this factor could act as a repressor of the transcription of these two SAUR genes. The results obtained in this thesis suggest that PAR1 might modulate development integrating light and hormonal signalling pathways.cat
