dc.description.abstract | [eng] The TGF-β pathway is currently considered a therapeutic target in advanced tumors, including glioblastoma (GBM), and several anti-TGFβ agents are in clinical trials and have shown promising results .A thorough understanding of the molecular mechanisms involved in the protumorigenic function of TGF-β will facilitate the identification of markers of response that can be used to stratify patients to be treated with anti-TGFβ compounds and, moreover, will allow for the discovery of new therapeutic agents. TGF-β is highly active in high-grade glioma, and elevated TGF-β activity confers poor prognosis. Aberrant activation of TGF-β is caused in part by enhanced secretion of the TGF-β ligands by tumor cells and tumor stroma cells. However, the precise mechanisms that lead to the hyperactivation of the TGF-β pathway are not yet fully understood. In this work, we have demonstrated that the TGF-β signaling pathway can be hyperactivated in GBM by different means including the stabilization of the TβRI by overexpression of the deubiquitinating enzyme USP15 or the enhanced secretion of TGF-β2 by the generation of a malignant autocrine loop via CREB. In this project, we identified USP15 and CREB as two novel regulators of TGF-β activity. USP15 and CREB could be considered as markers of response to anti–TGF-β molecules and potential therapeutic targets against GBM. | eng |
dc.description.abstract | [spa] La vía de señalización de TGF-β es una diana terapéutica en tumores avanzados tales como glioblastoma (GBM). Diversos fármacos anti-TGF-β están siendo evaluados en ensayos clínicos y muestran resultados esperanzadores. En GBM, la vía de señalización de TGF-β se encuentra hiperactivada y es factor de mal pronóstico ya que promueve invasividad, angiogénesis, metástasis, inmunosupresión e induce la auto-renovación de las “células iniciadoras de glioma”. La hiperactivación de la vía de señalización de TGF-β en GBM en parte está causada por un aumento de la secreción por parte de las células tumorales y las células del estroma de los ligandos de TGF-β. Sin embargo, los mecanismos precisos que originan la hiperactivación de la vía de señalización de TGF-β en GBM son aún ampliamente desconocidos. En este trabajo, hemos descrito dos nuevos mecanismos moleculares que dan lugar a una hiperactivación de la actividad TGF-β en GBM, tales como la estabilización de TβR-I debido a la sobre-expresión de la deubiquitinasa USP15 o la sobre-expresión de TGF-β2 debido a la generación de un ciclo maligno autocrino mediado por el factor de transcripción CREB. En este proyecto, hemos identificado USP15 y CREB como dos nuevos reguladores de la actividad TGF- β. Nuestros resultados sugieren que tanto USP15 como CREB podrían ser evaluados como dos nuevos marcadores de respuesta a fármacos anti-TGF-β y nuevas dianas terapéuticas contra GBM. | cat |
