Abstract: [spa] El objetivo principal de la Tesis ha sido profundizar en el conocimiento de la complejación de metales por ligandos de S y Se. Se ha dedicado una atención especial a las fitoquelatinas (PCn), que son la base de la fitorremediación, por su capacidad de complejar los metales pesados. Se ha intentado conocer los complejos formados, mediante experimentos realizados con diferentes técnicas analíticas. En una primera fase, el trabajo se centra en el uso de la polarografia de impulsos diferencial (DPP) asistida con el método quimiométrico de Resolución Multivariante de Curvas por Mínimos Cuadrados Alternados (MCR-ALS) para resolver las mezclas de péptidos presentes en plantas (fitoquelatinas y sus fragmentos) y Cd2+. Los resultados obtenidos se han comparado con los obtenidos por la espectrometría de masas (ESI-MS). El siguiente objetivo fue estudiar la fitoquelatina PC5, en presencia de los iones Cd2+ y Pb2+. El mayor tamaño de este péptido, respecto a otras PCn estudiadas anteriormente, podria implicar a priori modelos de complejación competitiva más complejos. Por ello, se planteó un estudio sistemático basado en una combinación del análisis quimiométrico (mediante MCR-ALS y los nuevos algoritmos pHfit y GPA) de las medidas por DPP y Dicroísmo Circular (CD), y ESI-MS. El tercer objetivo fue estudiar la selenocistina (SeCyst), en diversas situaciones de complejación competitiva. El comportamiento electrónico de SeCyst sola o en presencia de un ion metálico (Bi3+, Cd2+, Co2+, Cu2+ , Cr3+ , Ni2+ , Pb2+ y Zn2+) fue estudiado por DPP en un amplio intervalo de pH. Se han realizado también estudios por Valoración Isotérmica Calorimétrica (ITC) a pH fisiológico (7.4) que además, permitieron obtener parámetros termodinámicos y constante de complejación. En un estudio más detallado, se seleccionó el Cd2+ como modelo para comportamiento de SeCyst en la presencia de un metal no esencial, en cambio, se eligió el Zn2+ para el caso de un elemento esencial. Además, se estudiaran mezclas de SeCyst y el glutatión (GSH) en presencia de Cd2+ a pH fisiológico. Un último objetivo consistió en evaluar la posible utilización de electrodos modificados para la detección de tioles. En una estancia en la Universidad de Oxford, se llevaron a cabo estudios de modificación de dos tipos de electrodos de carbono: el de carbono vitrificado (GCE) y el de grafito pirolítico “edge plane pyrolytic graphite” (EPPG), inmovilizando en sus superficies un centro metálico redox (derivados del Ferroceno, Fc). Finalmente, se estudió su respuesta catalítica en presencia de Cys.
[eng] The main goal of the Thesis has been to deepen the knowledge about metal complexation by ligands containing S and Se. A special attention has been paid to phytochelatins (PCn), which are on the basis of phytoremediation due to their ability to complex heavy metals. The formed complexes have been studied by means of different analytical techniques. In a first step, the work is focused on the use of Differential Pulse Polarography (DPP), assisted by Multivariate Curve Resolution by Alternating Least Squares (MCR-ALS), in order to resolve the mixtures of peptides present in plants (phytochelatins and their fragments) and Cd2+. The obtained results have been compared with those by mass spectrometry (ESI-MS). The next goal has been the study of the phytochelatin PC5 in the presence of Cd2+ and Pb2+. The bigger size of this PCn as compared to other previously studied could imply a priori more complicated models of competitive complexation. Then, a systematic study was devised based on the combination of chemometric analysis (by means of MCR-ALS and the new algorithms pHfit and GPA) of the data obtained by DPP and circular dichroism, and ESI-MS. A third goal has been the study of selenocystine (SeCyst) in different situations of competitive complexation. The electrochemical behaviour of SeCyst alone or in the presence of a metal ion (Bi3+, Cd2+, Co2+, Cu2+, Cr3+, Ni2+, Pb2+ and Zn2+) has been studied by DPP in a wide pH range. Studies by isothermal titration calorimetry (ITC) have also been made, which have provided thermodynamic parameters and complexation constants. In a more detailed study, Cd2+ and Zn2+ ions have been selected as models of non-essential and essential metal, respectively, in order to study their behaviour in the presence of SeCyst. Mixtures of SeCyst and glutathione have been also studied in the presence of Cd2+ at physiological pH. The last goal has been to evaluate the possible use of modified electrodes for the detection of thiols. During a stay in the University of Oxford, several studies were carried out on the modification of two kinds of carbon electrode: glassy carbon (GCE) and edge plane pyrolitic graphite (EPPG). The modification consisted on the immobilisation of a metallic redox center (a ferrocene derivative) onto the surface. Then, the catalytic responses in the presence of Cys have been studied.
