Abstract: [spa] La presente tesis doctoral se ha realizado en el grupo de investigación de Catálisis Homogénea del Departament de Química Inorgànica de la Universitat de Barcelona. Este trabajo está dedicado a la síntesis y caracterización de nuevos ligandos quirales diamidofosfito, ligandos P-dadores que contienen dos enlaces P-N y un enlace P-O. Se han preparado 2 ligandos diamidofosfito monodentados y 16 bidentados. La síntesis se realiza en dos etapas por reacción de diaminas quirales enantiopuras ((R,R) y (S,S)-N,N’-dibencil-1,2-diaminociclohexano, (R,R)-N,N’-dimetil-1,2- diaminociclohexano y (R) y (S)-N,N'-dimetil-1,1'-binaftildiamina) y PCl3, seguido de alcoholes quirales ((2S,3S)-(+)2,3-butanodiol, (2R,3R)-(-)2,3-butanodiol, (1R,2R)-trans- 1,2ciclohexanodiol, (-)-2,3-O-isopropilideno-D-treitol, (S)-1,1’-bi-2-naftol y (R)-1,1’-Bi-2- naftol), también enantiopuros en presencia de una base. Los diamidofosfitos monodentados sintetizados son iónicos y contienen un grupo alcoxi funcionalizado con una sal de imidazolio. También se han sintetizado 6 diaminofosfinas bidentadas quirales a partir de diaminas enantiopuras ((R,R)-N,N’-dibencil-1,2- diaminociclohexano, (R,R)-N,N’-dimetil-1,2-diaminociclohexano y (R) y (S)-N,N'- dimetil-1,1'-binaftildiamina) y bis(diclorofosfinas) (1,1’-bis(diclorofosfino)ferroceno, 1,2- bis(diclorofosfino)etano y 1,2-bis(diclorofosfino)benceno), en presencia de una base. A partir de los nuevos ligandos se han preparado 31 precursores de Pd(II) del tipo: [Pd(η3-2-CH3C3H4)(P-P)]+, [Pd(η3-2-CH3C3H4)(P)2]+, [Pd(η3-1,3-Ph2C3H3)(P-P)]+ y [PdCl(η3-2-CH3C3H4)(P)] que se han evaluado en la reacción de alquilación y aminación alílica del rac-(E)-3-acetoxi-1,3-difenil-1-propeno con la sal sódica del malonato de dimetilo y bencilamina como nucleófilos. Se hace una comparación de los resultados de estas reacciones en disolvente orgánico y líquido iónico utilizando los precursores con ligandos diamidofosfito monodentados iónicos funcionalizados con una sal de imidazolio. Los mejores sistemas catalíticos en la reacción de alquilación y aminación en disolvente orgánico han resultado los precursores con ligandos con fragmento terminal derivado de la (R,R)-N,N’-dibencil-1,2-diaminociclohexano y fragmento puente derivado del butanodiol o del ferroceno. Con el fragmento terminal derivado de la N,N'-dimetil-1,1'-binaftildiamina son los que contienen el fragmento puente 1,1’-bi-2-naftol los que han dado mejores resultados. En líquido iónico el mejor precursor ha resultado ser el [PdCl(η3-2-CH3C3H4)P] con el ligando que tiene el heterociclo derivado de la (R)-N,N'-dimetil-1,1'-binaftildiamina y el grupo exocíclico derivado del tetrafluoroborato de 1-(2-hidroxietil)-3-metilimidazolio. También se han preparado 10 precursores de Rh(I) del tipo [Rh(COD)(P-P)]BF4 con una selección de los ligandos diamidofodfito y diaminofosfina y se han evaluado como precursores catalíticos en la reacción de hidrogenación de las olefinas proquirales modelo DMI, MAA y MAC. Algunos de los nuevos ligandos se han ensayado con los precursores de Rh(I) preparados “in situ”. En la reacción de hidrogenación el mejor precursor catalítico resulto ser el que contiene el ligando diamidofosfito bidentado sintetizado a partir de (R)-N,N'-dimetil-1,1'-binaftildiamina y fragmento puente (-)-2,3- O-isopropilideno-D-treitol, con el que se obtiene un 100% de conversión y un ee > 99% con los tres sustratos modelo. Este precursor también presentó buenas conversiones y enantioselectividades en la hidrogenación de enamidas cíclicas y Z-acetamidas. Por último se presentan los ensayos preliminares realizados en la reacción de hidroformilación del estireno utilizando el precursor de Rh(I) formado “in situ” donde los ligandos evaluados han presentado bajas actividades y enantioselectividades.
[eng]This Thesis work has been carried out in the Grup de Catàlisi Homogènia del Departament de Química Inorgànica de la Universitat de Barcelona. New enantiopure diamidophosphite ligands, which are P-donor ligands containing 2 P-N and 1 P-O bonds, have been synthesized. The ligands have a highly modular structure, which is well suited for the synthesis of small libraries of compounds. In total, two monodentate and sixteen bidentate diamidophosphite ligands have been isolated and characterized. The preparation of the ligands was accomplished by stepwise addition of optically pure substituted diamines (N,N’-dibenzylcyclohexane-1,2-diamine, N,N’- dimethylcyclohexane-1,2-diamine and N,N’-dimethyl-1,1’-binaphthyl-2,2’-diamine) and optically pure diols (butanediol, cyclohexanediol, di-O-isopropylidene-threitol and binaphthol) to phosphorus trichloride. The new monodentate ligands are ionic and contain an imidazolium salt functionalized with an alcoxy group. A series of six bidentate diaminophosphines have also been synthesized by reaction of enantiopure diamines (N,N’-dibenzylcyclohexane-1,2-diamine, N,N’-dimethylcyclohexane-1,2- diamine and N,N’-dimethyl-1,1’-binaphthyl-2,2’-diamine) with bis(dichlorophosphines) (1,1’-bis(dichlorophosphine)ferrocene, 1,2-bis(dichlorophosphine)ethane and 1,2- bis(dichlorophosphine)benzene). The corresponding bis(diamidophosphite) selenides were prepared and the 1JPSe were calculated in order to evaluate the sygma-donor ability of the new ligands. With the obtained ligands, 31 new Pd(II) complexes of the types [Pd(η3-2-CH3C3H4)(P-P)]+, [Pd(η3-2-CH3C3H4)(P)2]+, [Pd(η3-1,3-Ph2C3H3)(P-P)]+ and [PdCl(η3-2-CH3C3H4)(P)] have been prepared, characterized and tested in catalytic allylic alkylation and amination reactions using sodium dimethylmalonate and benzylamine respectively as nucleophiles in dichloromethane. The allylic substitution reactions have also been run in ionic liquid medium with Pd(II) precursors containing ionic ligands. The best results for the allylic substitution reaction in organic solvents have been obtained with Pd(II) precursors containing ligands with the terminal fragment derived from N,N’-dibenzylcyclohexane-1,2-diamine and the bridging fragments derived from butanediol or ferrocene. With complexes containing ligands with the terminal fragment derived from N,N’-dimethyl-1,1’-binaphthyl-2,2’-diamine those with binaphthol derived bridging fragment lead to the best results. In ionic liquid medium, complexes [PdCl(η3-2-CH3C3H4)(P)] with the ligand containing the heterocyclic fragment derived from N,N’-dimethyl-1,1’-binaphthyl-2,2’-diamine resulted to be the best precursor. Ten new [Rh(COD)(P-P)]BF4 complexes were synthesized with a selection of the new bis(diamidophosphite) ligands. The complexes were used as catalytic precursors in the asymmetric hydrogenation of benchmark substrates, namely methyl-alpha-acetamidoacrylate, methyl-(Z)-alpha-acetamidocinnamate and dimethyl itaconate. The influence of the nature of both the terminal and bridging fragments of the bis(diamidophosphite) ligands on the asymmetric induction is discussed. Most of the complexes proved to be effective catalytic precursors for the process. Total conversion and excellent enantioselectivity (>99% ee) in the hydrogenation of the three substrates was achieved with the complex containing the ligand with N,N’-dimethyl-1,1’-binaphthyl-2,2’-diamine derived terminal fragment and di-O-isopropylidene-threitol derived bridging fragment. The best performing catalytic precursor was tested in the hydrogenation of selected cyclic enamides and beta-acetamides. Preliminary tests in the asymmetric hydroformylation reaction of styrene have been run with “in situ” prepared Rh(I) precursors with some of the new ligands achieving low activity and enantioselectivity.
