Abstract: | [eng] AIMS: The general aim of this doctoral thesis is to improve current CAR-T immunotherapeutic strategies in the treatment of acute leukemias. The specific aims of this doctoral thesis are: 1. To develop and characterize CD123-directed CAR-DOT cells as an allogeneic-safe, off-the-shelf therapy far the treatment of R/R AML. 2. To engineer CD1axCD3 BiTE-secreting STAb-T cells and compare their effectiveness against conventional CD1a-directed CAR-T cells against coT-ALL. 3. To generate a CCR9-directed immunotherapy far the safe treatment of R/R T-ALL. (i) To validate CCR9 as an immunotherapeutic target far T-ALL. (ii) To generate CCR9-directed CAR-T cells. (iii) To generate dual CCR9/CD1 a-directed CAR. |
