[cat] Per tal de determinar el rol dels gens DOR i AIB3 en la diabetis de tipus 2 o en l'obesitat, es van fer dos estudis d'associació genètica amb mostres d'individus normotolerants i d'individus amb diabetis de tipus 2. Es van seleccionar diferents SNPs (un total de 15) localitzats en els dos gens i es van seleccionar 6 SNPs marcadors d'haplotip (htSNPs), segun criteris descrits en la bibliografia especialitzada. En total es van genotipar 542 individus control (dels quals 188 tenien un ÍMC inferior a 25) i 655 individus amb diabetis de tipus 2. Els resultats de l'estudi mostren associacions dèbils amb ÍMC per a dos polimorfismes localitzats en el gen AIB3 (p=0.021 y p=0.031). Igualment existeixen correlacions entre algun dels polimorfismes estudiats i paràmetres clínics en els individus no diabètics (n=214) com triglicèrids (p=0.010), i índex cadera-cintura (p=0.001). Aquests resultats suggereixen un possible rol d'aquests gens en la patologia de la diabetis l'obesitat, pel que es van iniciar diferents estudis funcionals. El primer, la generació d'un ratolí KO per a DOR; el segon l'estudi de l'expresió del gen en diferents patologies i en models animals. Els estudis en el ratolí KO hauran de permetre determinar el rol de DOR en diabetis i en obesitat. Pel que fa als estudis d'expressió s'observen diferències significatives en diferents situacions.
[eng] "Screening of susceptibility to type 2 diabetes: DOR and AIB3" To determine the role of DOR and AIB3 genes in type 2 diabetis or obesity, two genetic association studies were designed. To perform these studies, DNA samples from normoglucemic individuals and type 2 diabetic subjects were collected. Different SNPs (15) localized on the genes were selected, and following the guidelines published in specialized bibliography, 6 haplotype tagging SNPs (htSNPs) were selected to perform the association studies.Es van seleccionar diferents SNPs (un total de 15). 542 control subjects, and 655 diabetic subjects were genotyped for each polymorphism. The results of this study showed weak associations of two polymorphisms located on the AIB3 gene and BMI (p=0.021 y p=0.031). laso, some correlations with clinical data were observed within the control group. The results suggest a putative role of these genes in the pathology of type 2 diabetis and obesity. Following the genetic results some functional studies were done. First, the generation of a KO mouse for the DOR gene was started; and second, the expression of the genes DOR and AIB3 was evaluated in samples from control subjects and diabetic ones, and indeed in different animal models of pathology. The findings in these studies showed an strong regulation of the expression of the genes under some circumstances.
