Notas preliminares sobre las técnicas de amplificación y variación de la región ITS (Internal Transcribed Spacer) en el género Encalypta (Encalyptaceae, Bryophyta)

[spa] Se presentan las primeras aportaciones sobre el estudio molecular de la región ITS (Internal Transcribed Spacer), basado en la técnica de la PCR, para las especies del género Encalypta presentes en la Península Ibérica. El aislamiento del ADN se realizó a partir de fragmentos del gametófito y del esporófito procedentes tanto de material fresco como de herbario. También se ha utilizado protonema obtenido en cultivo in vitro, a partir de esporas. Los amplímeros obtenidos corresponden a las dos regiones espaciadoras (ITS-1 e ITS-2), además de la subunidad 5.8S, del ADNr nuclear. Se utilizaron dos protocolos de amplificación y las parejas de iniciadores ITS5/ITS4, ITS1/ITS4, ITS1/ITS2 e ITS3/ITS4. Con ITS5/ITS4 e ITS1/ITS4 se obtuvieron amplificaciones de la región ITS por completo, a partir del ADN extraido de material fresco. Sin embargo, a partir del material de herbario sólo se pudieron amplificar las regiones ITS-1 e ITS-2 por separado, utilizando los iniciadores ITS1/ITS2 e ITS3/ITS4.

[eng] Results on DNA amplifications are presented for the Encalypta species present in the Iberian Peninsula. DNA was isolated from the gametophyte and sporophyte, from both fresh and herbarium materials. DNA was also isolated from protonema obtained from cultures in vitro of spores. The internal transcribed spacers 1 and 2, including the 5.8S rnDNA have been amplified. Pair primers ITS5/ITS4, ITS1/ITS4, ITS1/ITS2 and ITS3/ITS4 were tested by using two different amplification protocols. ITS5/ITS4 and ITS1/ITS4 yielded amplimer only from fresh samples, not from herbarium collections, from which only primers ITS1/ITS2 and ITS3/ITS4 amplified successfully each of the ITS regions separately.
