[cat] La toxina epsilon de Clostridium perfringens és la toxina clostridial més potent després de les neurotoxines tetànica i botulínica. Provoca una enterotoxèmia general en animals de granja, provocant greus pèrdues econòmiques. La toxina es sintetitza en forma de protoxina inactiva que requereix de dos talls proteolítics per a la seva activació. En aquest treball hem clonat i expressat amb èxit en E. coli diferents formes de la protoxina i de la toxina, conjugades o no a la Green Fluorescent Protein (GFP), per el seu ús com a eina molecular d'estudi dels efectes de la toxina en els sistemes renal i nerviós de ratolí. Hem pogut demostrar que la toxina epsilon s'acumula en grans quantitats al ronyó, de manera inespecífica als túbuls proximals de la nefrona, i de manera específica als túbuls distals, on a més, provoca mort cel·lular de les seves cèl·lules epitelials. Pel que fa referència al cervell, hem pogut detectar acumulació específica tant de la protoxina com de la toxina epsilon a l'endoteli capil·lar arreu del cervell, i a més hem pogut comprovar que la toxina és capaç de travessar la Barrera Hemato-Encefàlica i acumular-se sobre cèl·lules de la glia neuronal. En estudis in vitro hem demostrat que la toxina és capaç de produïr mort cel·lular sobre cultius tant de cèl·lules endotelials com d'astroglia i microglia, suggerint que la seva acció sobre aquests tipus cel·lulars podria estar implicada en els efectes nerviosos que provoquen la mort última dels animals afectats.
[eng] "Clostridium perfringens" epsilon toxin is the most potent clostridial toxin after tetanus and botulinum neurotoxins. It causes general enterotoxemia in livestock leading to death, and is responsible for great economic losses every year. It is sintesized as a non-active prototoxin which needs txo proteolitic cleavages for becoming the mature active form of the toxin. In this study we have succesfully cloned and expressed in "E. coli" different recombinant forms of both the prototoxin and toxin, conjugated with Green Fluorescent Protein (GFP), for their use as a molecular tool in the study of epsilon toxin effects in mouse renal and nervous systems. We have demonstrated large accumulation of the toxin in the kidney. It accumulates in a non-specific way on the proximal tubules, and specifically in the luminal surface of the distal tubules of the nephron. It also causes the death of the epithelial cells of the distal tubules. On the brain, we detected specific accumulation of both the prototoxin and the toxin on the luminal surface of the endothelial cells. We found that the toxin is able to cross the Blood Brain Barrier and accumulate on glial cells. In vitro studies shown that the toxin causes the death of endothelial cell cultures and also of both astroglial and microglial cell cultures, suggesting that the action on these kind of cells could be implicated in the neural effects leading to affected animals death.
