dc.description.abstract[ENG] A collection of electron micrographs illustrating details of the seminiferous tubules, spermatogenic cells, and spermatozoa in the rat testis. Towards the year 2010, an online virtual microscope project led by Dr Xavier Messeguer (Universitat Politècnica de Catalunya) and Dr Fernando Aguado (Universitat de Barcelona) was launched and hosted at www.genomedu.com. The project was discontinued in 2021 or 2022, and the material became unavailable. Some of the electron microscopy images I contributed at that time have now been adjusted for brightness and contrast, with lettering and labelling added, and figure legends prepared for inclusion in this collection. Unfortunately, it was not possible to examine the original grids and sections again, and some elements could not be identified or photographed at higher resolution. When processing those samples, rats were perfused with a mixture of formaldehyde and glutaraldehyde, postfixed in osmium tetroxide and maleate-buffered uranyl, and embedded in EPON 812. Thin sections approximately 50 nm thick were counter-stained with uranyl acetate and lead citrate. Cap al 2010, el Dr. Xavier Messeguer (Universitat Politècnica de Catalunya) i el Dr. Fernando Aguado (Universitat de Barcelona) van liderar un projecte de microscopi virtual en línia accessible a www.genomedu.com. El projecte es va cancel·lar en 2021 o 2022 i el material va deixar d'estar disponible. Algunes de les imatges que vaig aportar al projecte han estat ara ajustades de brillantor i contrast, s'han afegit lletres i símbols, s'han redactat uns peus de figura i s'han inclòs en aquesta col·lecció. Per a l'obtenció d'aquestes mostres, les rates es van perfondre amb una barreja de formaldehid i glutaraldehid, es postfixaren en tetraòxid d'osmi i uranil en amortidor maleat, i s'inclogueren en EPON 812. Els talls ultrafins, d'uns 50 nm de gruix, es van contrastar amb acetat d'uranil i citrat de plom.

Seminiferous tubule in a rat with adjacent interstitial tissue. The tubule is enclosed by a thick basement membrane and a layer of myoid cells with flattened nuclei (my). The nucleus of Sertoli cells (Se) is ovoid, indented, and oriented perpendicular or parallel to the basement membrane. Small, round nuclei in this zone were tentatively identified as dark type A spermatogonia (Ad). Large nuclei located at mid-levels within this specialised stratified epithelium are likely to be primary spermatocytes (1s). At more superficial levels, spermatids can be seen at various stages of maturation: Golgi phase (G), during which multiple Golgi complexes and proacrosomal granules gather together; cap phase (c), when the acrosomal vesicle forms a cap over the anterior half of the nucleus; acrosome phase (a), during which the nucleus becomes condensed and elongates; and maturation phase, where the compacted head of sperm cells (s) is distinguished. Portions of the lumen (L), representing extracellular space, can be seen between cells at some distance from the surface. In the interstitial tissue, a small arteriole (ar), a fibroblast (fi), and what appears to be a dendritic cell (den) with numerous cell processes are observed. The nuclear envelope in the anterior part of the spermatids thickens (ant, white arrowheads), and an incipient acrosomal vesicle appears in two cells (black arrowheads). The heads of late spermatids or spermatozoa (heads) at late stages of spermiogenesis – acrosome and maturation phases – are also observed. Spermatids at various stages of spermiogenesis in rats. Golgi complexes and proacrosomal granules are grouped (G), and the nuclear envelope thickens (ant, white arrowheads) in this anterior part of the cell. At the opposite pole of the cell – and laterally – mitochondria (m) lie peripherally beneath the cell membrane. Cells in late phases of spermiogenesis are visible, with compacted spermatozoon heads (heads) that have not been detached yet from the residual portions of cytoplasm (residual). Transverse sections at various levels of spermatozoon tails are present, including thin end pieces (end). Epoxy section ≈50 nm thick; uranyl acetate and lead citrate counterstaining.
