La cultura de trabajar sin prisa, sin pausa y sin nervios. 7. ¿Qué se debe hacer para tener una entrevista de trabajo con éxito? 8. Calidad factor común de las personas con éxito. 9.     The SeDeM expert system is a methodology which is applied in preformulation and formulation studies of medicines specifically in solid dosage forms. This system informs on the physical profile of powdered substances (APIs and excipients) used to formulate drugs (Suñé et al, 2005; García et al, 2010; Aguilar et al, 2009). By determining whether powders (API or excipient) are suitable for direct compression, the SeDeM profile will inform about the advantages and gaps of those powdered substance to be used in direct compression, so the system informs on whether the direct compression method is appropriate (e.g.. wet granulation should be applied before compression). The characterization of powdered substances by SeDeM facilitates the identification of the characteristics that require amendment in order to obtain tablets by direct compression. This system thus provides information that will ensure the robust design of the formulation in the final product.     An overview on the modulation of the intestinal barrier and immune response by membrane vesicles secreted by the probiotic Escherichia coli Nissle 1917

    Probiotic Escherichia coli Nissle 1917 (EcN) is a good colonizer of the human gut and its efficacy in the inflammatory process undergone in ulcerative colitis has been demonstrated. The probiotic action is mainly through the modulation of intestinal epithelial tight junctions and immune system. Here we review the role of outer membrane vesicles (OMVs) released by this probiotic strain on the modulation of intestinal homeostasis. EcN OMVs enter into host epithelial cells via clathrin-mediated endocytosis and are sorted to lysosomes via endocytic compartments. In cellular models of intestinal barrier, EcN OMVs stimulate the underlying immune system through the intestinal epithelium, triggering immune and defense responses. Thus, the use of probiotic derived OMVs could be a safe probiotic-derived strategy targeting intestinal inflammatory processes.

    Magnetic nanoparticles: From diagnosis to therapy

    Magnetic nanoparticles have proven to be promising theranostic agents, namely tools for therapy and diagnosis. Among them, superparamagnetic iron oxide nanoparticles (SPIONs) highlight for their biocompatibility and reduced toxicity. Here, we describe the synthesis and characterization of SPIONs by co-precipitation of ferric and ferrous salts under mild conditions. These particles were able to accumulate in inflamed areas fact that was increased upon the application of an external magnetic field. Resonance magnetic imaging studies have shown their suitability as negative contrast agents for diagnosis. In addition, hybrid nanoparticles were obtained by incorporating the above described SPIONs into liposomes or nanoemulsions. The findings have confirmed the high potential of these systems for biomedical applications.

    The lipid-protein interplay in efflux plump

    In this work lactose permease (LacY) of Escherichia coli has been taken as model efflux pump to investigate the interactions between the protein and the main lipid components (POPE and POPG) of the inner membrane. Two main approaches have been followed: (i) measuring the fluorescence energy transfer between a single tryptophan mutant of the protein (W151/C154G LacY) and pyrene labeled phospholipids (Pyr-PE and Pyr-PG); and (ii) pulling the protein from the supported lipid bilayers where it is embedded by using the tip of the atomic force microscope (AFM). On one hand, fluorescence measurements at different pHs indicate that LacY present selectivity for PE. On the other hand the observations of the reconstituted protein in lipid bilayers by AFM show a preference of LacY for the fluid phase (Lα) rather than for the gel phase (Lβ). To get an estimation of the proportion of each lipid in each phase we have constructed a phase diagram for the system POPE:POPG. The diagram shows that at the temperature of the experiments (24 ºC) there is an almost equimolar proportion of each lipid. The results suggest the existence of a boundary region around LacY formed mainly by POPE laterally segregated from a bulk with a random distribution of POPE and POPG. Force spectroscopy allows to establish the force required and the mechanism to unspecifically unfold the protein.

    A multicompartment compliance aid (MCA) is a blister-type repackaging system that aims to facilitate drug administration and thereby increase patient adherence. One of the characteristics of the MCA that should be taken into account is the moisture permeability, since this atmospheric condition is one of the most important factors that can modify the stability of medicines. In the current paper we report the moisture permeability tests performed on a MCA according to the US Pharmacopeia. This information on the suitability of the device will help pharmacists implement a high-quality professional service.

    Protein misfolding and aggregation into amyloid-like structures is related with an increasing number of both non-neuropathic (either localized or systemic) and neurodegenerative human disorders. Decrypting the mechanisms and implications underlying amyloid assemblies has become a central issue in biology and medicine. Compelling evidence show that the formation of amyloid aggregates has a negative impact in cell physiology, entailing the cell dysfunction and finally apoptosis and cell death. The aim of the present review is to illustrate the currently status of the most common and/or debilitating conformational diseases, from Alzheimer to prion diseases.

    Passport to a profession in the degree of pharmacy: how to integrate university students into the real world of work: "Passport to a profession in Pharmacy"

    The incorporation of the Spanish university system into the European Higher Education Area has brought about a series of adaptations. Among the recommendations is the inclusion of an external training period in a company, which has resulted in significant changes in the degree syllabus in order to balance the theoretical and practical education required by the students. This new framework has been legally confirmed by the Spanish Government and, in the case of the University of Barcelona, by the publication of internal guidelines. Taking advantage of this new opportunity to adapt the Pharmacy degree to real-world problems in industry, the Dean’s team of the Faculty of Pharmacy, with the support of the Faculty administrative staff and the Students Advisory Service, have assumed the challenge of including a new subject in the syllabus entitled Training in Companies. In parallel, a new activity has been set up to ensure that the students choose the most suitable company department/job for them and to help them pass the company interview. Under the name of Passport to a Profession, a series of ten explanatory talks has been scheduled every academic year. These talks deal with a broad range of topics aimed at providing the students with the basic tools they will need to make the most of a company training period and to make headway in the professional world when they finish their degree. In addition, three Faculty of Pharmacy-Pharmaceutical company workshops and two round-table conferences have been held in the last two years in order to bring the university and industry together. Notably, the project to provide students with company training is expanding on an international level, with two to three undergraduate students contracted every year by a United Kingdom-based multinational pharmaceutical company. The statistical data of the whole process has been analysed for a more in-depth understanding of the activity and to improve the programme.

    Membrane interaction of polymyxin B and synthetic analogues studied in biomimetic systems: implications for antibacterial action

    Membrane-active antimicrobial peptides, such as polymyxin B (PxB), are currently in the spotlight as potential candidates to overcome bacterial resistance. We have designed synthetic analogs of PxB in order to determine the structural requirements for membrane action. Since the mechanism of action of PxB involves interaction with both the outer membrane and the cytoplasmic membrane of Gram negative bacteria, we have used an approach based on mimicking the outer layers of these membranes using monolayers, Langmuir-Blodgett films and unilamelar vesicles, and applying a battery of biophysical methods in order to dissect the different events of membrane interaction. Collectively, results indicate that the PxB analogues act in the bacterial membrane by the same mechanism than PxB, and that cationic amphipathicity determines peptide activity.

    Clinical pharmacokinetics of mycophenolic acid and its metabolites in solid organ transplant recipient

    Mycophenolate mofetil (MMF), an ester prodrug of the immunosuppressant mycophenolic acid (MPA), is widely used for maintenance immunosuppressive therapy and prevention of renal allograft rejection in renal transplant recipients. MPA inhibits inosine monophosphate dehydrogenase (IMPDH), an enzyme involved in the “de novo” synthesis of purine nucleotides, thus suppressing both T-cell and B-cell proliferation. MPA shows a complex pharmacokinetics with considerable interand intra- patient by between- and within patient variabilities associated to MPA exposure. Several factors may contribute to it. The pharmacokinetic modeling according to the population pharmacokinetic approach with the non-linear mixed effects models has shown to be a powerful tool to describe the relationships between MMF doses and the MPA exposures and also to identify potential predictive patients’ demographic and clinical characteristics for dose tailoring during the post-transplant immunosuppresive treatment.

    Bicelles: new nano systems for skin applications

    Bicellar systems are lipid nanostructures formed by long- and short-chained phospholipids dispersed in aqueous solution. Because of their attractive combination of lipid composition, small size and morphological versatility, bicelles became new targets for skin research. Bicelles modify the skin biophysical parameters and modulate the skin barrier function acting as enhancers for drug penetration. Moreover, these aggregates have the ability to penetrate through the narrow intercellular spaces of the skin stratum corneum and to reinforce its lipid lamellae. Their structures allows for the incorporation of different molecules that can be carried through the skin layers. The remarkable versatility of bicelles is their most important characteristic, which makes it possible their use in different fields. These aggregates represent new nanosystems for skin applications. In this work we provide an overview of the main properties of bicelles and their effects on the skin.
